Potassium Fluxes in Dialyzed Squid Axons
Measurements have been made of K influx in squid giant axons under internal solute control by dialysis. With [ATP](i) = 1 microM, [Na](i) = 0, K influx was 6 +/- 0.6 pmole/cm(2) sec; an increase to [ATP](i) = 4 mM gave an influx of 8 +/- 0.5 pmole/cm(2) sec, while [ATP](i) 4, [Na](i) 80 gave a K influx of 19 +/- 0.7 pmole/cm(2) sec (all measurements at approximately 16 degrees C). Strophanthidi...
متن کاملRemoval of Potassium Negative Resistance in Perfused Squid Giant Axons
Squid giant axons, internally and externally perfused with solutions having potassium as the only cation, exhibit an approximately linear steady-state current-voltage relation. When small amounts of calcium and magnesium are present in the external potassium solution, the current-voltage curve is markedly nonlinear, exhibiting the rectification and negative resistance which have been observed f...
متن کاملActivation-inactivation of potassium channels and development of the potassium-channel spike in internally perfused squid giant axons
A spike that is the result of calcium permeability through potassium channels was separated from the action potential is squid giant axons internally perfused with a 30 mM NaF solution and bathed in a 100 mM CaCl2 solution by blocking sodium channels with tetrodotoxin. Currents through potassium channels were studied under voltage clamp. The records showed a clear voltage-dependent inactivation...
متن کاملTracer and Nontracer Potassium Fluxes in Squid Giant Axons and the Effects of Changes in External Potassium Concentration and Membrane Potential
The efflux of labeled and unlabeled potassium ions from the squid giant axon has been measured under a variety of experimental conditions. Axons soaked in sea water containing 42K ions lost radioactivity when placed in inactive sea water according to kinetics which indicate the presence of at least two cellular compartments. A rapidly equilibrating superficial compartment, probably the Schwann ...
متن کاملDivalent cations and the activation kinetics of potassium channels in squid giant axons
The effects of external Zn+2 and other divalent cations on K channels in squid giant axons were studied. At low concentration (2 mM) Zn+2 slows opening kinetics without affecting closing kinetics. Higher concentrations (5-40 mM) progressively slow opening and speed channel closing to a lesser degree. In terms of "shifts," opening kinetics are strongly shifted to the right on the voltage axis, a...
